Cultivation strategies for prokaryotes from extreme environments

چکیده
The great majority of microorganisms are as-yet-uncultivated, mostly found in extreme environments. High-throughput sequencing provides data-rich genomes from single-cell and metagenomic techniques, which has enabled researchers to obtain a glimpse the unexpected genetic diversity “microbial dark matter.” However, cultivating environments remains essential for dissecting utilizing functions extremophiles. Here, we provide straightforward protocol efficiently isolating prokaryotic different habitats (thermal, xeric, saline, alkaline, acidic, cryogenic environments), was established through previous successful work our long-term experience extremophile resource mining. We propose common processes isolation at first then summarize multiple cultivation strategies recovering meanwhile specific tips that always overlooked but important. Furthermore, use multi-omics-guided microbial approaches culturing these as-yet-uncultivated two examples provided introduce how work. In summary, allows significantly improve efficiency pure cultures novel taxa, therefore paves way protection utilization resources

Viruses from extreme thermal environments.
Viruses of extreme thermophiles are of great interest because they serve as model systems for understanding the biochemistry and molecular biology required for life at high temperatures. In this work, we report the discovery, isolation, and preliminary characterization of viruses and virus-like particles from extreme thermal acidic environments (70-92 degrees C, pH 1.0-4.5) found in Yellowstone...
متن کاملMagnetotactic Bacteria from Extreme Environments
Magnetotactic bacteria (MTB) represent a diverse collection of motile prokaryotes that biomineralize intracellular, membrane-bounded, tens-of-nanometer-sized crystals of a magnetic mineral called magnetosomes. Magnetosome minerals consist of either magnetite (Fe3O4) or greigite (Fe3S4) and cause cells to align along the Earth's geomagnetic field lines as they swim, a trait called magnetotaxis. ...
متن کاملEditorial: Enzymes from Extreme Environments
Enzymes are nature's biocatalysts and are equipped with high catalytic activity and remarkable substrate specificity. Enzymes catalyze the metabolic reactions necessary for all of life's processes and are able to catalyze reactions at much higher rates than would be possible without their action. The majority of enzymes are optimized to perform under physiological conditions or conditions consi...
متن کاملClose Interspecies Interactions between Prokaryotes from Sulfureous Environments
Green sulfur bacteria are obligate photolithoautotrophs that require highly reducing conditions for growth and can utilize only a very limited number of carbon substrates. These bacteria thus inhabit a very narrow ecologic niche. However, several green sulfur bacteria have overcome the limits of immobility by entering into a symbiosis with motile Betaproteobacteria in a type of multicellular as...
متن کاملExtreme catalysts from low-temperature environments.
Cold-loving or psychrophilic organisms are widely distributed in nature as a large part of the earth's surface is at temperatures around 0 degrees C. To maintain metabolic rates and to prosper in cold environments, these extremophilic organisms have developed a vast array of adaptations. One main adaptive strategy developed in order to cope with the reduction of chemical reaction rates induced ...
